چکیده

Patterned microarrays of antibodies were fabricated and tested for their ability to bind targeted bacteria. These arrays were used in a series of bacterial immunoassays to detect E. coli 0157:H7 and Renibacterium Salmoninarum (RS). Microarrays were fabricated using microcontact printing (μCP) and characterized using scanning probe microscopy (SPM). The high-resolution SPM imaging showed that targeted bacteria had a higher binding selectivity to complementary antibody patterns than to unfunctionalized regions of the substrate. Additional studies indicated a significant reduction in binding of bacteria when the microarrays were exposed to non-specific bacteria. These studies demonstrate how protein microarrays could be developed into useful platforms for sensing microorganisms.

On-chip microbial culture for the specific detection of very low levels of bacteria.

Microbial culture continues to be the most common protocol for bacterial detection and identification in medicine and agronomics. Using this process may take days to identify a specific pathogen for most bacterial strains. Surface Plasmon Resonance (SPR) detection is an emerging alternative technology that can be used for the detection of bacteria using protein microarrays although typical limi...

The Design of Simple Bacterial Microarrays: Development towards Immobilizing Single Living Bacteria on Predefined Micro-Sized Spots on Patterned Surfaces

In this paper we demonstrate a procedure for preparing bacterial arrays that is fast, easy, and applicable in a standard molecular biology laboratory. Microcontact printing is used to deposit chemicals promoting bacterial adherence in predefined positions on glass surfaces coated with polymers known for their resistance to bacterial adhesion. Highly ordered arrays of immobilized bacteria were o...

Single-cell-based sensors and synchrotron FTIR spectroscopy: a hybrid system towards bacterial detection.

Microarrays of single macrophage cell-based sensors were developed and demonstrated for potential real-time bacterium detection by synchrotron FTIR microscopy. The cells were patterned on gold electrodes of silicon oxide substrates by a surface engineering technique, in which the gold electrodes were immobilized with fibronectin to mediate cell adhesion and the silicon oxide background was pass...

Microarrays for bacterial detection and microbial community analysis.

Several types of microarrays have recently been developed and evaluated for bacterial detection and microbial community analysis. These studies demonstrated that specific, sensitive and quantitative detection could be obtained with both functional gene arrays and community genome arrays. Although single-base mismatch can be differentiated with phylogenetic oligonucleotide arrays, reliable speci...

Biostickers: Patterned Microfluidic Stickers for Rapid Integration with Microarrays

We present a one-step, reversible, and biocompatible bonding method of a stiff patterned microfluidic "Biosticker", based on off-stoichiometry thiol-ene (OSTE) polymers [1], to state-of-the-art spotted microarray surfaces. The method aims at improving and simplifying the batch back-end processing of microarrays. We illustrate its ease of use in two applications: a high sensitivity flow-through ...
